1. Preparation of Fmoc-Lys(Boc)-resin
1) Take 100g of 2-chloro-triphenyl chlororesin (0.5mmol/g resin, 50mmol), soak in 1000ml DMF for 30 minutes, make the resin fully swollen, add DIPEA 20ml, Fmoc-Lys(Boc)-OH 23.5g, 25°C for 65 minutes. Add 50ml of methanol and react at 25°C for 60 minutes.

Semaglutide Growth Trajectory:
In December 2017, the U.S. Food and Drug Administration (FDA) officially approved semaglutide injection for blood sugar control in adults with type 2 diabetes.
In September 2019, the US FDA approved Rybelsus (oral semaglutide) for medical use.
In January 2020, the FDA granted its new use approval again to reduce the risk of major adverse cardiovascular events in adult patients with type 2 diabetes mellitus with cardiovascular disease. This product is a monoglycosylated form of bovine enterokinase light chain, recombinant expression by Pichia yeast and purified by high-performance chromatography, containing 6 His-tags, theoretical molecular weight of about 43 kDa, optimal digestion pH of 4.5-9.5, and optimal digestion temperature of 25°C. Recombinant interic kinase activity is inhibited to varying degrees by imidazole, sodium chloride, glycerin, phosphate, etc.
